It depends on whether you drag the folder itself to the Desktop, or a link to the folder. In the latter case, deleting the link will not affect the folder itself. In the former case, instead of deleting the folder, drag it back to its original location (or wherever you want it to be).

It depends on various factors, such as the specific Desktop Environment and the File Manager that you're using. You could look for documentation on those, or experiment. Holding a meta key, such as Shift or Control, sometimes affects the way dragging works.
You can also accomplish it with shell commands, if you're familiar with those.
