From a rescue boot you must chroot to the installed system. From there, with most distros, you can run passwd to set a new root prompt. In others it may set a new password for a sudoer. I'm not personally familiar with the latter, because I have no sudoers in *buntu or Mint or any of their derivatives that default to having no root login configured.
