I would suggest posting separate problems separately in respective forum categories (Hardware, for the Nvidia question). You might get better results.
That said, wine has to be executed on an .exe file. So for example, let's assume you wanted to install something from a CD, say Photoshop. You would insert the CD, "cd" in to the CDROM ("cd /mnt/cdrom") and run the .exe setup program with:
wine setup.exe
You will find that many linux/unix tools run are this way.
If you prefer, you can also set wine as the default application for .exe files (most windowm mangaers, KDE, GNOME support this), and then just double click on them.
