I need to start do development for a web application using PHP connected to MYSQL DB on Linux Ubuntu Desktop PC,Can Please advice the best way to how can I start with linux OS.

Use a Live distribution that boots from a CD/DVD/USB to try it out. Many of them will let you very easily mount existing disk partitions to access the data in your installed operating system.
what do you really mean by "start with linux os" ? Could you be more specific?
If you're afraid of working on linux, then don't think about it: grab a live distro and see what happens. I would suggest you knoppix. There are ways to prevent the distro to write anything to your hard drive, so you won't be able at all to destroy your data or something like that.
If you already have installed ubuntu on your desktop pc, then install emacs or eclipse and you're done.
See turnkey linux for a pre-made virtual machine that has what you want. There are other places too that offer all the software installed in some disto's or pre-made vm's or builds. SuseStudio could create what you want but as a newbie you may wish to avoid any installing and use a pre-made lamp system.
You can also rent a linux box from a provider. Rackspace is a very popular one. I use digitalocean, it costs $5 a month.
This is the quickest way to get linux up and running without having to worry about equipment or compatibility. Also you are able to test sites as they would be seen to actual customers without having to set up firewall rules and messing with your router or cable modem.
