I think what you're asking has little to do with media playback, and everything with filesystem reads? I.e., how do file read speeds differ between NTFS and ext4? Or did you have some other "Linux native file system" in mind? They might also perform differently.
Generally speaking, these reads
must always be significantly faster than actual file playback. I'm not sure what sort of "inefficiency" you have in mind there.
A slow USB connection could be a much more significant bottleneck.
