Mono is a good option. But it lacks microsoft proprietry libraries (which might be an issue).
If you don't have anything you really want to keep on wine, then trying the winetools might be a wise idea. Its supposed to help you set up your wine environment correctly. But I have found it doesn't play well with a basic configuration that may have evolved if you installed wine with debian. And I have experienced and seen in this forum people having problems with various aspects.
If you don't have any existing code (or can easily port it to non-ms specific .NET) then I'd recommend you check out the mono.NET project. It would make more sense, if you have professional development in mind.
