I have just spent several hours trying to get it working on my AMD64 system.
Highlights:
The gtk2 gui requires librsvg. This dependency is completely undocumented.
Without that library installed, no error message. An executable "mlgui" is generated without any gui functionality.
When this executable is started, it generates over 20 configuration files in current work directory. If that happens to be your home directory or some other place with important files in it, happy cleaning. It also creates ~/.mldonkey directory but doesn't seem to use it for anything useful.
After installing all required libraries and recompiling mldonkey, the gui starts. However, trying to do anything with it causes a segmentation fault.
I like the idea of one application supporting all different networks, but mldonkey implementation is bad.
