Backup Server MySQL (very big)

I have a MySQL server with more 100GB database, currently i am using replication for backup.
I run on Centos OS, MySQL latest version.
But i have some problems with replication, i want to every body consultation for some backup solutions ?

What's exactly the problem while using mysql replication for your backup?
What engines does your DB using? I suggest create a script and then dump your database and use remote copying utility to store your backup DB.

No need to reinvent the wheel

Have a look at this great script at SourceForge: AutoMySQLBackup. I use it on production servers to take hourly, daily, weekly and monthly backups and it works like a charm.
