Doesn't your bootloader come with instructions on how to achieve this?

I was thinking more like eliminating the ability for users to choose anything other than what comes up on the menu.

You can password protect the grub shell and individual menu items.

Also check the "S" entry in inittab:
Code:
              # What to do in single-user mode
              ls:S:wait:/etc/init.d/rc S
              ~~:S:respawn:/sbin/sulogin
sulogin requires the root password.
See the grub info file (section 9) and the inittab man page.

If the lab computer can be booted from CD or USB or floppy, they could boot up, running a live distro and access files that way.

Like I hinted at before, this should be covered in your bootloader's documentation. If you want to see if someone can provide you with a quick summary of the steps involved, you'll need to at least tell us what bootloader you are using.
