Then just go ahead and install it on that partition. I don't know what distro you'll want to install but if it's a polished and mature distro I guess that the installation procedure will dump you into some form of partition manager, where you decide what partition or partitions to use, what file systems and where to mount it/them. Just point out the new partition and choose it to be formated with a file system (ext3 is a good bet), mounted on / and off you go. At the end of the installation process you'll probably get a question about grub and booting. Again, this varies between distros but if it's a polished and mature distro so I'd be surprised if it doesn't autodetect Knoppix and ask you if you want the grub boot menu to contain both distro and to write it to the MBR.
What is the other distro? Do you have any other OS on the computer?
