Can I setup Fedora, OpenSuse, and Ubuntu as a print server in medium size company. The company consist of 250 staffs. Should i have to acquire the license?

Yes, of course, you'll want to check out http://cups.org but what license? there is no license to buy. This isn't Windows.

please use better thread titles in future. this is a Linux forum, so "Linux" isn't that useful... :)

The license is free of charge. It only tells that if you modify the source code, your modifications will be as free as the original code in case you decide to distribute it. You don't need to modify the code to setup a print server, therefore the license should not be a problem. So there is no license to acquire since you're not distributing anything.
You may consider contracting for support though. This can save your business in case of trouble.
