Why do you want this? Not in the way you expect it. Linux has a predefined directory structure and files must be in the correct place (e.g. libraries go to /lib). There are some programs which comes witout a package (e.g. Java, here you can decide to install it e.g. in /opt/java if you like it). RPM and deb packages copy the files always in the predefined place. But you can mount a partition at any plce in the directory tree, so you can use a separate partition for /usr/share, /var, /opt and so on. To get an overview of the Linux directory tree and what goes where have a look at

yes, you can

You can have anything on any hard disk you want, provided that:

- the hard disk has a file system that is supported by the current kernel
- the hard disk or any of it's partitions is mounted somewhere in the Linux file tree

Read the man pages to fdisk and mnt or ask a clear question on what you want achieve exactly.
