You can change config file. It will not activate your new configuration until you restart or reload

service squid reload
$SQUID $SQUID_OPTS -k reconfigure

service squid restart
basically service stop and start

I had compiled squid from source and use following command to refresh squid.conf configuration changes while squid keep running:

/usr/local/squid/sbin/squid -k reconfigure

If you installed from an rpm then please verify your squid path and then issue the switch "-k reconfigure" just as i had typed above.
