When you add or delete a route, it adds or deletes the route in the kernel routing table. You can display the current routing table with a 'route' command. ( without quotes ). Network Manager is just a front end to modify the routing table. You can add or delete routes with 'route add 111.222.333.444' for example. What goes in the ifcfg-eth* files is static information you usually do not have a need to change on a day to day basis. ( no routing information though ).
