Will Dell OptiPlex 5050 Desktop QUADCORE i5, 3.2GHz 16GB DDR4 2TB hdd run Linux?

It will indeed run linux. It may take a bit of work to get all of the features working.
Try some live media.
Many of the top distro's offer live media and they don't even say it.
Try Clear Linux OS as it is basically built by Intel for speed. It's dvd is live and a good working test disc.

Yes. I have had a number of Dells over the years, and they have all worked quite nicely with Linux.
The only thing you need to check out is the wireless chipset, if you are planning to use wireless.

Everything worked "out of the box" on my older Dell (laptop) except the Broadcom wireless. But that problem is easily resolved. If the Broadcom 43xx chipset is being used, search for "no_net_install_bcm43xx_firmware" tar archive. (Comes up as the first hit on DDG. Worked like a charm for openSUSE. Would likely work on other flavors.)
