My server is currently running RHEL 5. I would like to install RHEL 6 over it. Do I simply install it and will the installation erase everything I have on the server now?
when you try to install it, it will detected the current file system layout and ask you to either use it or format the partitions. I would recommend that you format the partitions to ensure a clean installation. There is no upgrade path from Major released e.g 5 to 6. Also the default filesystem is now ext4. I would suggest ensuring your backups are good and verified and perform an clean installation, erasing all disk information in the process.
To add to the above good advice; RH does NOT support in-place upgrades across major version nums eg 5.x => 6.x, and for very good reasons.
Backup all your stuff and make notes, then do a fresh install of v6. https://access.redhat.com/kb/docs/DOC-35284
