If your machines are not networked the only option you have is to tranfer the file to an wxternal disk and physically move from one to the other

Yes, data transfer with RS323 is possible, but very slow. It also would be pointless, you would need a cable for that and if you have to buy it you can just get a network cable instead.
Other option: Remove the disk from one machine, connect it with the other machine, copy the data.

this would be the best way to handle it. either network the computers or physically move the HDDs around and use DD or some other tool to move the data about internally.

The best and more practical way is to networked the computers.
Other ways is by copying the file(s) to a USB/SD drive or burning the files to a DVDRW disc and repeating the tedious process until all file(s) are copied.

Without knowing the capabilities or specifications of these computers it is a bit difficult to help but basically you can transfer data using just about any port.
Serial port (RS-232)
Parallel Port
USB (Requires a special purchased cable)
Firewire (IEEE 1394)
As already stated using serial port and parallel will be very slow compared to a LAN.
