Newer software, new fixes for the new software, new security patches for the new software..that's it mainly. Maybe some old programs removed and/or replaced with newer one.

The difference is basically the same than between RHEL4 and 3, or between any other two RHEL versions, or any two Fedora (Core) versions. It's always the same story: new software, new version. Sometimes (but not necessarily now) there comes along something really new, like "we just dropped using ext2 only and jumped into the LVM world!". Or "hey, now we have a graphical front-end for installing new packages!".

Read RHEL official news.
