BTW, I'm yet to test Ventoy with Fedora.

In a online search I was not able to find anything in regards to this.

But on this Ventoy page it states it worked with some Fedora flavors / versions, none of them Fedora 33 editions, though.

It does not mention persistence either.

My guess is Fedora does not require a specific label to create the dat file, so it will default to casper-rw.

Will post back here with results when done.

I have used MANY distribution ISOs with E2B and fewer with Ventoy only because it is newer. At no time has it ever been important what Distro or OS I created the USB key FROM, only what ISO I copied to the USB device. That said, I have only tested a few with Persistence, and no version of Fedora. (Which, I am sorry, has never impressed me much for most purposes.)

I would suggest you use the method described in post 4 at the link below. You need to extract or loop mount the Kubuntu iso to a directory and then copy all the contents to the vfat partition on the usb drive and follow the other steps. If you are doing this from fedora, make sure you have the i386-pc directory in the fedora /boot/grub directory. If you don't, copy it there from /usr/lib/grub before running grub-install and make sure you get the correct drive.

If you want persistence on the Kubuntu usb, use gparted or another partition manager to create a second ext4 (or other Linux fs) partition and label it casper-rw. When this is done, go to the /boot/grub/grub.cfg file and at the end of the kernel line (the line beginning with linux) you should see quiet splash --. Hit the space bar and type in the word persistent and save.
