When I check the setting in the GNOME sound manager I get the following error:
audiotestsrc wave=sine freq=512 ! audioconvert ! audioresample ! gconfaudiosink profile=chat: Could not open audio device for playback. Device is being used by another application.
It's been a while for me, so you'll have to forgive me for forgetting to mention it, but after you run alsaconf and alsamixer run alsactl store as root.

The alsactl store saves the settings for next boot.
