It is trivial to serve DNS for more than one subnet, the tricky part is DHCP services for two subnets.

What divides the two subnets?

Being primarily a network guy these days, I might make sure we have one or more routers physically between, and provide a DNSMasq server on each subnet.

I guess the full answer to your question is just "yes", but that does not sound helpful enough. Can you provide a little more information?
