Few questions about postfix!!!

Hello All,
In Postfix mail server the mail queue directory is /var/spool/postfix. Now I have around 100 users in my current setup.
and server is giving the error like no more space is avialable. When I run du --max - =1 /var/spool/postfix as super user,
I see that it has occupied plenty of space. there is no more space avialable in /var directory which is mounted on /dev/sda4. SO now I need to delete the them . Shall I run #postfix flush ?????????? as
superuser.

But I fail to understand one thing that all mail users are residing in /home directory and their mailbox is in /home/user/MailDir
and plenty more place is leaft in /home which is mounted on /dev/sda3.
Gurus plese explain this?
