Is it possible to create a custom font.conf file

If it is possible, what are parts that should be included in it?

Do you mean such as in /etc/fonts? Sure -- here's a local.conf file that I use:
prompt: cat /etc/fonts/local/conf
<?xml version="1.0"?>
<!DOCTYPE fontconfig SYSTEM "fonts.dtd">
<!-- /etc/fonts/local.conf file to configure system font access -->
