what options did you use in your command?
Assuming your iso file is located in your home directory
and its name is puppy.iso

mount -o loop puppy.iso ISO/puppy

should normaly mount as read and write. If not you can add
the 'rw' option:

mount -o loop,rw puppy.iso ISO/puppy

PS
I just checked on my system (slackware12.0)

# mount -o loop slampp-2.0.1-zw-dvd.iso /mnt/tmp
# ls -l /mnt/tmp
total 9
-rwxr-xr-x 1 root root 55 2009-08-12 18:22 autorun.inf
drwxr-xr-x 6 root root 2048 2009-08-12 18:21 boot
drwxr-xr-x 9 root root 2048 2009-08-12 18:22 slampp-2.0-zw
-rwxr-xr-x 1 root root 4286 2009-08-12 18:22 slampp.ico

As you can see, it's read and write for the owner

There might be some individual files or/and folders that are
read-only but you can change that with the chmod command

Of course all of this has to be done as root
