Hi Knoppixguy
I have the CD version of Knoppix 4.0.2. My USB hard drive is sda1.
First the ISO image was copied to a USB drive (sda1).
In theory, just use the boot option (also called "cheat code"):
knoppix tohd=/dev/sda1.
Subsequent boots are done with the option:
knoppix fromhd=/dev/sda1
Subsequent boots require the CD to bring up the boot screen. However, the "fromhd" boot option runs Knoppix from the ISO on the USB drive.
For me, there were some problems, but in the end it worked.
On my PC, the process of copying the ISO to the USB hung up before completion. The actual process of getting the entire ISO to the USB drive was clumsy.
And yes, live distros are much more fun to run from a USB drive than from the CD/DVD.
Good Luck!
