New entries to /etc/modeprobe.conf

If I add new entries to /etc/modprobe.conf, do I have to reinstall the kernel for them to be picked up in /boot/initrd.xxx.xx.xx.xx.img?

No need to reinstall kernel or rebuild as long as the module is already present in the working kernel. If it is not there in the kernel then it must be compiled and added.
