You will have to mount the windows partition yourself. There are a number of guides on how to do this about - note that if your Windows partition is NTFS then you will need to install an ntfs kernel module.

Samba is for sharing files on a Windows network, if you only need local access then you don't need samba (from the way your question was phrased it sounded like this was what you were needing to do). If you do need to set it up over a network, you will have to give it the location where you mounted the windows partition yourself. Again, there are a number of guides out on the net on how to do this.

I still don't exactly understand what you need. Are you saying that on your local computer (i.e. not on a network), you want your Windows partition to be automatically mounted on boot up? If this is the case, you don't need samba. As I stated in my previous post, samba is only for sharing/accessing files over a Windows network.

To automatically mount a Windows partition on boot, all you need to do is modify your /etc/fstab. There are a lot of sites that give you the general outline of how to do this, for example here. Note that you may need to install an ntfs kernel module if your Windows partition is formatted as ntfs.

If this is not what you need, ask again, but please be clear what you're trying to do!
